RNA-Interferenz ist ein naturlicher Mechanismus des Organismus zur posttranslationalen Genexpressionsregulierung, kann aber auch therapeutisch genutzt werden. SiRNA-basierte Therapeutika sind vor allem in der Behandlung von Krebs, viralen und genetischen Erkrankungen vielversprechende Entwicklungen, die neue Gen-Angriffspunkte bieten. Einige Vertreter befinden sich bereits in klinischer Testung. Allerdings ist die Anwendungsmoeglichkeit siRNA-basierter Therapie beschrankt. Eines der groessten Probleme ist die geringe Zellaufnahme. Um diese zu verbessern werden unterschiedliche chemische Modifikationen, Konjugate und Nanopartikel entwickelt. In dieser Arbeit wurden Konjugate von Designed Ankyrin Repeat Proteinen (DARPins) mit siRNA hergestellt. DARPins sind Proteine, die spezifisch Antigene binden koennen und Antikoerpern in vielen Faktoren uberlegen sind. DARPin-siRNA Konjugate koennten neue Strategien fur eine Target-spezifische RNAi-basierende Therapie ermoeglichen.
