In Cars Race-O-Rama folgen die Spieler den Abenteuern von Lightning McQueen und Chick Hicks im neuen Stadion von Radiator Springs. In der eigenen Akademie bereiten die beiden ihre Schüler auf die Race-O-Rama-Rennserie vor. Die Spieler begegnen 12 neuen spielbaren Charakteren, die in Cars Race-O-Rama ihr Videospieldebüt feiern. Lightning McQueen wurde mit speziellen Ersatzteilen stark verbessert und kann so noch schneller und eleganter über die brandneuen und actiongeladenen Strecken rasen. Hierbei treten die Spieler auf den Straßen einer Großstadt, in der Baja-Wüste und auf sandigen Strandabschnitten an. Zusätzlich können jetzt Lightning McQueens Reifen, Felgen, Motorhaube, Seitenschweller, Heckspoiler und Wagenfarbe mit zahlreichen Möglichkeiten verändert werden. So erhält Lightning ein einzigartiges Aussehen, das es so noch nie zuvor gegeben hat. Die Spieler können in diesem Rennabenteuer zudem Lightning McQueens erweiterte Driftfähigkeiten nutzen und so "Kachow"-Posen für die begeisterten Reporter und Fans im Spiel ausführen.
